摘要
目的 探讨苯乙酸钠 (Na PA)诱导人前列腺癌细胞株 (PC- 3)凋亡及其分子机理。方法 应用流式细胞术 (FCM)观察不同剂量 Na PA对 PC- 3细胞周期改变的影响 ,通过 RT- PCR检测 bcl- 2和增殖细胞核抗原 (PCNA)在 m RNA水平的表达。结果 PC- 3细胞体外经不同剂量 (0m M/ L,1 .5m M/ L,3.0 m M/ L,6.0 m M/ L) Na PA处理后 ,细胞周期的 G0 / G1 期升高 ,S期、G2 / M期相对下降 ,G0 / G1 峰前有明显的亚二倍体峰 ,即凋亡峰 ,且随着 Na PA浓度的增加 ,凋亡细胞数逐渐增多 (P<0 .0 0 1 ) ;bcl- 2和 PCNA在 m RNA水平随 Na PA剂量的增加 ,表达量下降。结论 Na PA可能通过下调 bcl- 2和 PCNA在 m RNA水平的表达诱导 PC- 3细胞凋亡。
Objective To study apoptosis and its molecular mechanism induced by sodium phenylacetate (NaPA) in human prostate carcinoma cell strain (PC-3).Methods The effects of NaPA with different dosages (0 mM/L;1.5 mM/L; 3.0 mM/L; 6.0 mM/L) on the biological changes of apoptosis and cell cycle in PC-3 were observed by flow cytometry technique (FCM). The mRNA expression level of bcl-2 and PCNA was detected by RT-PCR.Results PC-3 cell growth was inhibited and apoptosis was induced by NaPA in a dose-dependent manner. The G 0/G 1 phase cells significantly increased and S or G 2/M phase cells relatively decreased afer the treatment.The bcl-2 and PCNA mRNA expression levels decreased with dosage-dependent of NaPA increasing.Conclusions The PC-3 cell apoptosis induced by NaPA may be regulated by decreasing the expression of bcl-2 and PCNA on the mRNA level.
