摘要
目的 建立OxLDL诱发大鼠血管内皮细胞凋亡的模型。方法 SD大鼠尾静脉注射非氧化的LDL ,2 4h后取主动脉血管内皮细胞铺片 ,采用光学显微镜和荧光显微镜进行形态学观察 ,TUNEL法染色计算凋亡细胞比例。结果 ( 1)静脉注射LDL组观察到明显的凋亡形态学改变 ,对照组未见凋亡内皮细胞 ;( 2 )静脉注射LDL ( 4mg/kg、 6mg/kg、8mg/kg)组凋亡细胞比例分别为 8 10 %、 18 92 %、 2 2 0 3 % ,三组间有非常显著性差异 (P <0 0 1)。结论 ( 1)尾静脉注射LDL后 2 4h可引起血管内皮细胞凋亡 ;( 2 )
Objective To establish a rat model to study endothelial cell (EC) apoptosis induced by oxidized low density lipoprotein (OxLDL) in vivo.Method Sprague-Dawley rats were injected in the tail vein with unmodified human LDL. 24 hours after the injection, samples of the aortas were taken to prepare 'en-face' specimens of aortic endothelium. EC apoptosis was detected by morphological analysis under light and fluorescence microscopy. TUNEL technique was used to estimate the ratios of EC apoptosis. Result (1)The typical apoptotic morphological changes were found in the groups which were injected with LDL but not in the control groups which were injected with PBS. (2)The apoptotic ratios of the groups (4mg/kg, 6mg/kg and 8mg/kg, IV) were 8.01%,18.92%and 22.03%, respectively. The differences between the 3 groups were significant(P<0.01). Conclusion (1)EC apoptosis can be induced at 24 hours after the injection of LDL. (2)Intravenous injection of LDL evokes EC apoptosis in a dose-dependent manner.
