摘要
目的 建立更加简便实用的类风湿关节炎模型 ,为类风湿关节炎发病机制的研究提供良好的实验材料 ;观察盘状结构域受体 2 (DiscoidinDomainReceptor2 ,DDR2 )在类风湿模型动物早期的表达 ,为探讨DDR2与类风湿关节炎滑膜细胞损伤的关系提供依据。方法 放射影像学、酶联免疫检测和免疫组织化学。结果 从病理学、影像学、血清学及临床特征观察模型的改良情况 ,符合类风湿关节炎临床特征的个体达到 85 %以上 ,且建模时间明显缩短 ;免疫组化显示 ,DDR2免疫反应阳性产物定位于关节囊滑膜细胞和滑膜下层细胞的胞膜和胞浆。结论 经改良的佐剂型类风湿关节炎模型优于常规的动物模型 ;
Objective To establish simpler and more practicable animal models for the research on rheumatoid arthritis(RA) and provide more satisfactory material for the study on the pathogenesis of RA ; to observe the expression of Discoidin DomainReceptor 2(DDR2) in the early phase of RA model and do the groundwork for deeper study. Method Radiology , enzyme-linked immunoabsorption assay and Immunohistochemistry. Result According to pathological, radiological and serological examinetions as well as to the rats' general status and clinical situation, more than 85% of the individual met the requirements. Furthermore, the episodetime was reduce prominently. The Immunohistochemicalresults demonstrated that DDR2 was expressed not only in the synovial cell line on the surface of synovial tissues of the articular capsule, but also in many other cells under the synovial line, and the positive signal mainly existed in the cytoplasm and cell membrane.Conclusion Under these experimental conditions ,we can establish animal models for RA easilier and faster; DDR2 can express peculiarly during the earlier phases of RA.
