摘要
目的 :为了从海洋无脊椎动物中寻找生物活性物质 ,对山东青岛及其附近海域常见的 2 1种海洋无脊椎动物的粗提物进行了免疫调节活性测定。方法 :测定了它们的乙醇提取物以及不同极性部位对T、B淋巴细胞增殖的调节作用。在小鼠脾细胞体外培养中用T细胞丝裂原ConA(伴刀豆蛋白A)和B细胞丝裂原LPS(脂多糖 )分别刺激T、B细胞增殖 ,以DNA合成前体3H TdR(胸腺嘧啶核苷 )掺入法检测T、B淋巴细胞的增殖强度。结果 :海洋无脊椎动物对T、B淋巴细胞具有ConA或LPS协同刺激作用或者抑制作用 ,其中 10种有明显的剂量依赖性 ,6种呈现小剂量增殖、大剂量抑制的双向调节作用。结论 :海洋无脊椎动物粗提物具有一定的免疫调节功能 ,这些数据为从海洋无脊椎动物中寻找天然药物活性前体奠定了基础。
Objective:The immunomodulatory effects of the ethanol extract of 21 marine invertebrate species were determined.Methods:The ConA and LPS were used to activate the proliferation of T and B lymphocytes in mice spleen in vitro.Then the precursor of DNA synthesis 3H-TdR was added to indicate the extent of proliferation.Results:It was found that the ethanol extracts of the marine invertebrates have immunomodulatory activity on the proliferation of T and B lymphocyte.Among them,10 showed well concentration-dependent immunomodulatory effect,and 6 have exhibited fine dual-immunomodulatory effects at the doses of 1~100 μg/ml.Conclusion:The results suggested that the marine invertebrates have some immunomodulatory effects and deserved further investigation.These study are the base to find new lead compounds in marine invertebrates.
