摘要
目的 :为了探讨IL 13基因编码区精氨酸 (Arg) 110谷氨酰胺 (Gln)多态性是否与湖北地区汉族人群哮喘及血浆总IgE水平升高相关。方法 :采用PCR RFLP方法 ,检测湖北地区 4 3名哮喘患儿、4 5名成人哮喘患者、31名非哮喘儿童和 4 6名体检健康成人的IL 13外显子 4Arg110Gln的等位基因频率和基因型频率。用化学发光法测定血浆总IgE。结果 :IL 13基因4 2 5 7位等位基因a在儿童、成人中的频率分别为 0 39、0 32。IL 13基因Arg110Gln多态性的GlnGln型与儿童哮喘和血浆总IgE升高相关 (P分别为 0 0 30、0 0 0 0 9) ,但与成人哮喘、血浆总IgE水平之间无统计学意义 (P分别为 0 2 19、0 174 )。结论 :研究显示IL 13外显子 4Arg110Glng a单核苷酸多态性与湖北汉族儿童哮喘和血浆总IgE水平相关 ,但与成人哮喘、血浆总IgE水平之间无统计学意义。
Objective:To investigate whether Arg110Gln polymorphism in the coding region of the IL-13 gene is associated with asthma and total plasma IgE level in Han nationality in Hubei Chinese population.Methods:Using restriction fragment length polymorphism to detect the allele frequency of 4257(g/a) site and Arg110Gln genotype of IL-13 in Han nationality in Hubei Chinese population,which included 43 asthmatic children,45 asthmatic adults,31 control children and 46 control adults. Total plasma IgE was measured by Chemiluminescence assay.Results:The frequency of allele a at 4 257 bp of IL-13 of children and adults was 0.39 and 0.32, respectively.The GlnGln form of Arg110Gln polymorphism of IL-13 gene was associated with susceptibility of asthma and elevated total plasma IgE in children( P =0.030 and 0.000 9, respectively),but not with them in adults( P =0.219 and 0.174, respectively).Conclusion:The results suggest that the Arg110Gln polymorphism of IL-13 gene is associated with susceptibility of asthma and elevated total plasma IgE in Chinese children of Han nationality in Hubei,but not with them in adults.
