摘要
目的 探讨AQP_4基因表达与缺血再灌注损伤后脑水肿之间的关系。方法 插线法制作大鼠脑缺血再灌注损伤模型,测脑组织含水量。原位杂交法测定缺血再灌注后6、12、24、48、72h AQP_4 mRNA在脑组织中的表达变化。结果 缺血再灌注后视上核、下丘脑、海马、大脑皮层等部位AQP_4 mRNA表达上调,24~72h达高峰。脑组织含水量亦在48~72h达高峰。结论 随缺血再灌注后脑水肿的出现,AQP_4 mRNA表达上调。这一结果提示AQP_4在缺血再灌注后脑水肿的形成中可能发挥着重要作用。
Objective To study the relationship between AQP_4 mRNA expression and brain edema after cerebral ischemic reperfusion
(CIR). Methods CIR models were established by occluding unilateral middle cerebral arteries(MCA) of rats with ropes for 2 hs and
reperfusing them for 6~72 hs. The expression of AQP_4 mRNA was measured by in situ hybridization. Brain edema was measured by
percentage of hemispheric enlargement at the same time. Results The expression of AQP_4 mRNA was up-regulated in supraoptic nuclei,
hypothalamic nuclei, hippocampus, cerebral cortex and other regions, and reached a peak at 24~72 hs after CIR. The water content in
ischemic brain tissue reached its peak at 48~72 hs after CIR. COnclusion The expression of AQP_4 mRNA was up-regulated following the
brain edema. The results implied that AQP_4 might play an important role in the formation of brain edema after CIR.
