醋酸泼尼松对家兔硫唑嘌呤药代动力学的影响

THE PHARMACOKINETIC EFFECT OF PREDNISONE ON AZATHIOPRINE IN RABBITS

目的：证实醋酸泼尼松和硫唑嘌呤合用是否合理，为临床应用提供实验依据。方法：采用反相高效液相色谱法对单用硫唑嘌呤及醋酸泼尼松与硫唑嘌呤合用后，对家兔体内硫唑嘌呤的代谢产物六巯基嘌呤（６ － ＭＰ） 的血清药物浓度进行了测定，并对两组药物动力学参数进行配对ｔ 检验统计处理。结果：单用硫唑嘌呤及两药合用后硫唑嘌呤的代谢产物６ －ＭＰ的药代动力学参数（ ＡＵＣ、Ｃｌ 、Ｔｐｅａｋ 、Ｃｍａｘ 、Ｔ１／２ａ 、Ｖｃ 、Ｔ１／２ｋａ 、Ｔ１／２β ）无显著性差异。结论： 醋酸泼尼松与硫唑嘌呤可以联合应用。

AIM To confirm whether it was suitable to use prednison(Pred) with Azathioprine(AZP) and provide experimental data for clinical use.METHOD The serum concentrations of AZP metabolite 6-Mercapto prine(6-MP) in different time were determined by reverse phase HPLC,when AZP was used alone and combined with Pred in nine rabbits.The pharmacokinetic parameters of the two groups wer statistically analysed by paired t test.RESULTSThe pharamacokinetic parameters(AUC,CL,T peak ,C max ,T 1/2a 、V c 、T 1/2ka 、T 1/2γ 、V c 、T 1/2β ,)of 6-MP were not significantlys different in two groups.CONCLUSION Azathioprion and Prednison can be used in combination.
