摘要
目的:为寻求特异性强、灵敏度高的一氧化氮测定方法,解决体内一氧化氮测定的难题。方法:用N- 甲基- D-葡糖胺二硫代甲酸铁[(MGD)2Fe2+] 作为一氧化氮的自旋捕集剂,对小鼠肝、心、肾和肺脏中一氧化氮水平进行电子自旋共振(ESR) 测量。结果:对1-0μmol·L- 1的一氧化氮水平,用自旋捕集方法可得到明显的ESR信号;对生物组织可测到1-0×10- 9mol·mg-1 的一氧化氮。
AIM In order to find a special and sensitive method to measure NO and to solve the difficulties of probing NO in vivo.METHODS The ferrous N-methy-D-glucamine dithiocarbamate[(MGD) 2Fe 2+ ] was used as NO spin trap to probe the NO quantity in liver and heart and kidney and lung of mice.RESULTS ESR signal is obvious when NO concentration is 1 0μmol·L -1 ,the least testable quantity of NO in tissue is 1 0×10 -9 mol·mg -1 .CONCLUSION A high sensitive and distinctive method was established to measure NO in situ in biological system.
