摘要
目的:观察Cl- 通道阻断剂呋喃苯胺酸对重组人白细胞介素-1β(interleukin- 1β,IL- 1β)诱导的小鼠骨髓基质细胞(BMSC) 增殖作用的影响。方法:MTT法测定原代培养的小鼠BMSC增殖活性。结果:IL-1β(200~1 000kU·L-1) 对BMSC具显著的促增殖作用。呋喃苯胺酸(20 ~500μmol·L-1)特异性地抑制IL- 1β的促增殖作用,效应呈浓度依赖性。结论:Cl- 通道与IL- 1β促BMSC增殖作用有关,Cl-
AIM To study the effect of chloride channel blocker furosemide on recombinant human interleukin-1β (IL-1β)-induced proliferation of mice bone marrow stromal cells (BMSC).METHODS The growth of primary cultured BMSC was determined by MTT assay.RESULTS IL-1β at the concentration of 200~1 000kU·L -1 stimulate the proliferation of BMSC significantly,and furosemide at the concentration of 20~500μmol·L -1 specially inhibited the effect of IL-1β in a dose-dependent manner.CONCLUSION Chloride channels play an important role in the signal transduction of IL-1β in bone marrow stromal cells
