高效液相色谱法测定鱼腥草中槲皮素的含量

DETERMINATION OF QUERCETINUM IN HOUTTUYNIA CORDATA THUNB BY HPLC

目的 :建立鱼腥草中槲皮素的反相高效液相色谱法 ,并用该法对不同产地鱼腥草中槲皮素的含量进行测定。方法 :采用AlltimaC18硅胶柱 ,以乙腈 - 0 .4 %磷酸 ( 4 5 :5 5 )为流动相 ;3 70nm波长处检测 ;流速 :1.0ml·min-1;柱温 :3 0℃。结果 :线性范围为 0 .2 4～ 1.2 0 μg ,相关系数 r =0 .9996,平均回收率为 96.3 0 % ,RSD =1.19%。结论 :该方法简便、准确 。

AIM To develop a HPLC method for determination of quercetinum and study the difference of Houttuynia cordata Thunb from different districts.METHODS Quercetinum was determined by HPLC on Alltima C 18 column 250mm(4.6mm,5μm) at 30℃ with acetonitrile-0.4% phosphoric acid ( 45∶55) as the mobile phase.Detection wavelength was 370nm. The flow rate was 1.0ml/min.RESULTS The calibration curve of quercetinum was 0.24～1.20μg(r=0.999 6).The average recovery of quercetinum were 96.30%(RSD=1.19%).CONCLUSION The method was simple and accurate and apt to control the quality of Houttuynia cordata Thunb.
