大鼠黑质的衰老变化——Ⅱ.乙酰胆碱酯酶活性药物组织化学法和图像分析系统定量研究 被引量:2

摘要 本研究用药物组织化学方法,对6只老年大鼠和5只成年大鼠黑质神经元的乙酰胆碱酯酶(AChE)活性进行了对比研究,并用图像分析仪对黑质 AChE 阳性神经元数目、胞体 AChE 活性及细胞面积进行了定量和比较。本文的结果表明,老年大鼠黑质 AChE 阳性细胞数量非常显著减少,细胞重新合成 AChE 的速率和量都显著减低、灰度值增高,AChE 细胞胞体的面积也随年龄缩小。本文在动物或人脑细胞衰老变化的研究中,增添了新的资料。老年时黑质神经细胞明显变性,必定使纹状体多巴胺(DA)能和胆碱(ACh)能系统之间平衡失常,致使老年的运动功能协调出现障碍。 By means of pharmacohistochemical regimen method and image analysis system, the age-related changes of acetylcholinesterase activity of the substantia nigra neu- rons and the number and size(section area)of the AChE positive neurons in rats were investigated quantitatively.The results showed that the number of AChE pos- itive neurons in the substantia nigra of the old rats decreased markedly,the rate and quantity of AChE synthesis in the perikaryon remarkable declined.The size (section area)of the AChE positive neurons also reduced with aging.These changes with aging have never been studied in human or usual experimental animals,so that some new parameters were provided for the research field in experimental geronto- logy. In human and animal,the degeneration of the neurons in substantia nigra cau- sed by aging would disturb the balance between dopaminergic and acetylcholinergic system and hence interfere with the normal coordination of movement.
