非抗病毒治疗相关的慢性乙型肝炎患者YMDD变异研究

Study of YMDD mutations in the patients of chronic hepatitis B untreated with Killing virus's medicines

目的 :了解未经抗病毒治疗的慢性乙型肝炎患者乙型肝炎病毒基因 YMDD变异及其与前 C区变异、HBV基因型的相关性 ,HBV DNA水平与变异发生的关系。方法 :从 1 2 6例均未接受拉米夫定治疗和近一年内未经任何抗病毒治疗的慢性乙肝患者随机抽取 2 5例为研究对象。血清 YMDD变异检测运用微板核酸杂交 -核酸定量法 ,HBV基因型采用 PCR微板核酸杂交 - EL ISA技术 ,HBV DNA定量采用荧光定量 PCR分析系统 ,HBV- M采用 EL ISA法。结果 :7例存在 YMDD变异 ,变异者 HBe Ag全部阳性 ,不同的 HBV基因型形式存在不同的 YMDD变异率 ,HBV DNA水平与 YMDD变异的发生不呈正相关结论 :HBV存在 YMDD野生变异株 ,并非完全由抗病毒治疗引起 ,变异株与前 C区变异和 HBV基因型的相关性以及

Objective:To explore the relation between the YMDD mutations and it's pre C domain mutation and genotypes of hepatitis B virus(HBV),also to investigate the relationship of level of HBV DNA and possibility of mutations in the patients of chronic hepatitis B untreated with Killing virus's medicine Methods:The twenty five cases optional by 126 patients of chronic hepatitis B untreated with lamivudine and Killing virus's medicine one year were detected.The YMDD mutations of serum was detected by microplate sandwich hybridization nucleic acid ration,genotypes of HBV was detected by PCR microplate sandwich hybridization ELISA,HBV DNA quantifying assessment was detected by fluorescence ration PCR analysis,hepatitis B virus markers were detected by ELISA Results:Mutations were detected in seven of 25 sample,all mutation's HBeAg were positive. Mutations rate is not same in each genotype Conclusion:There are some YMDD mutations in the patients of chronic hepatitis B untreated with Killing virus's medicine.It need to farther research about the relationship of YMDD mutations and pre C domain mutation and genotypes of HBV
