摘要
目的 :了解肺癌转移过程中基因的参与 ,进而为肺癌诊断、治疗提供依据。方法 :选用 5个不同转移潜能的体外培养肺癌细胞株H1 2 99、HLAMP、PGCL3、PAa和NiS作为研究对象 ,以永生化的支气管上皮细胞 1 6HBE为对照 ,采用含有 80 0个基因探针的肺癌相关基因芯片LUs ,研究各细胞株基因表达的差异。结果 :5个肺癌细胞株差异表达的基因共有 35 5个 ,5株细胞共同上调或下调的基因仅有 4个 ,基因表达的变化在不同细胞株之间明显表现出多态性。其中 4个转移株与无转移的NiS间比较 ,有 9个基因表达差异 ;3个高转移细胞株H1 2 99、HLAMP、PGCL3与和低转移细胞株PAa和无转移NiS之间比较 ,有 1 3个基因表达差异。这些基因大多是膜蛋白、细胞骨架蛋白 ,或和细胞信号转导有关 ,基因功能复习提示和肿瘤转移有可能的关联性。结果还显示和细胞膜陷窝结构有关的蛋白可能在肺癌转移过程中具有较大的意义。结论 :基因芯片方法对于筛选肺癌转移相关基因有独特的优势 ;获得的差异表达基因具有较强的代表性 。
Objective: To investigate the involvement of gene in the process of lung cancer metastasis,hence providing evidences and data for designing the early diagnosis and therapy protocols. Method: 5 cell strains of different sources and metastasis potentials were used to compare the differential expression of 800 genes arrayed in Lus cDNA chips. The lung cancer cell strains used in this experiment were H1299,HLAMP,PAa,PGCL3 and NiS,and 16HBE as control. Results: There were totally 355 genes of 5 tumor cell strains differentially expressed in comparison with the genes in 16HBE cell. Only 4 genes showed the same expression tendency,and polymorphism of gene expression was presented among all the 5 lung cancer strains. It was observed that there were 9 genes with altered expression when comparing the four stains with metastasis potential with NiS stain which does not have metastasis potential;13 genes with altered expression when comparing 3 cell strains presenting high metastasis potential with the 2 cell strains with low metastasis and without metastasis character. Study of these gene functions suggested their possible roles associated with lung cancer metastasis. Conclusion: DNA chip method is a powerful way to screen the gene associated with lung cancer cell metastasis with high representativeness. Further investigation of these positive genes may significantly facilitate the research of better understanding the mechanism of lung cancer metastasis.
