摘要
目的 :观察诱生型一氧化氮合酶基因在心肌梗死后大鼠早期的表达变化。方法 :将大鼠 36只随机分为心肌梗死组和假手术组 ,心肌梗死组又分为 1、4、8、1 2、2 4h组 ,用RT PCR检测各组心肌iNOSmRNA的表达。结果 :假手术组心肌无iNOSmRNA的表达 ,心肌梗死后 1h心肌组织即有iNOSmRNA的表达 ,8h、1 2h达到高峰 ,随后下降 ,8、1 2h和 1h相比差异显著 (P <0 .0 5 )。结论 :正常心肌组织无iNOSmRNA的表达 。
Objective: To observe the inducible nitric oxide synthase gene expression in the early stage of myocardial infarction in rats. Method: 36 rats were randomly selected and divided into the group of myocardial infarction and the group of sham operation,while the group of myocardial infarction was subdivided into groups of 1,4,8,12,and 24h after myocardial infarction. The iNOS gene expression was assessed using reverse transcription-polymerase chain reaction (RT PCR) standardized with β action. Result: There was no iNOS mRNA expression in sham operations,mild iNOS mRNA expression one hour after myocardial infarction,which peaked at hours 8 and12,then declined thereafter. Significant difference was noted in iNOS mRNA expression between hour 1 and hours 8 and 12. Conclusion: There were no expression of iNOS mRNA in the normal myocardium,there were expression of iNOS mRNA in the early stage of myocardial infarction.
