前列腺特异膜抗原mRNA检测方法的建立及应用

摘要
目的 建立前列腺特异膜抗原 (PSM)mRNA检测方法。方法 利用异硫氰酸胍 /酚 /氯仿 /异戊醇一步法从前列腺癌患者外周血有核细胞或前列腺癌组织中提取总RNA ,进行RT PCR扩增。结果 在PCR反应体系中投入相当于 1～ 1 0 0个癌细胞所含总RNA量时 ,均能得到清晰的 2 6 2bp的产物带。结论 本法有很高的稳定性和灵敏度。临床应用有助于早期发现前列腺癌转移。
