摘要
目的:研究转化生长因子β1(TGF-β1)、α平滑肌肌动蛋白(α-SMA)在慢性胰腺炎组织中的表达及其临床病理意义。方法:应用免疫组织化学EnVisionTM法,检测TGF-β1、α-SMA在38例慢性胰腺炎及20例正常胰腺组织中的表达。结果:TGF-β1在慢性胰腺炎组织中主要表达于胰腺星状细胞(PSC)、腺泡细胞及导管上皮细胞的胞质中,而在正常胰腺组织的腺泡细胞及导管上皮细胞中则不表达;α-SMA在慢性胰腺炎组织中主要表达于胰腺血管或导管平滑肌细胞、PSC的胞质中,而在对照组的PSC中几乎不表达。TGF-β1、α-SMA的表达强度,在正常胰腺组织中要明显弱于慢性胰腺炎组织(P<0.01),并且它们的表达强度与胰腺纤维化的程度呈正相关(P<0.01)。结论:TGF-β1是慢性胰腺炎胰腺纤维化过程中的关键因子,而PSC则是产生细胞外基质(ECM)的主要细胞;抑制TGF-β1的表达和PSC的活化在抗胰腺纤维化的治疗方面有着较好的前景。
Objective:To study the expression of transforming growth factor-β1 (TGF-β1 and a-smooth muscle actin(a-SMA) in chronic pancreatitis (CP) tissues and its clinical pathological significance. Methods:EnVision immunohistochemical staining was used to detect the expression of TGF-β1 and a-SMA in 38 cases of CP patients and 20 normal pancreatic tissues. Results:The expression of TGF-β1 in CP was mainly seen in pancreatic stellate cells (PSC), adenocytes and ductal cells. No TGF-β1 expression was found in normal pancreatic adenocytes and ductal cells. a-SMA in CP was mainly expressed in pancreatic vascular or ductal smooth muscle cells and cytoplasm of PSC. The expression of a-SMA was not found in normal pancreatic stellate cells. The expression of TGF-β1 and a-SMA in normal pancreatic tissues was obviously weaker than that in CP (P<0. 01). The degree of pancreatic fibrosis was related to their expression intensity (P<0. 01). Conclusion:TGF-β1 is a key factor in the course of pancreatic fibrosis and PSC is the main cell that produce extracellular matrix (ECM). It will be very useful to inhibit the expression of TGF-β1, and activation of PSC in the treatment of pancreatic fibrosis.
