人胃癌组织中环氧化酶-2 mRNA的表达 被引量:1

Expression of cyclooxygenase-2 gene in human gastric neoplasmas

摘要 目的 :检测环氧化酶 2 (COX 2 )mRNA在人胃癌、癌旁及正常胃黏膜组织中的表达情况 ,探讨其表达与胃癌临床病理特征的关系。方法 :应用逆转录 聚合酶链反应 (RT PCR)技术检测 2 4例胃癌、癌旁及正常胃黏膜组织中COX 2mRNA表达水平。结果 :COX 2mRNA在胃癌组织中的表达水平为 1.2 0 8± 0 .412 ,高于其在癌旁组织中的 0 .5 0 4± 0 .10 7和正常胃黏膜组织中的 0 .473± 0 .0 98,差异具有显著性 (P <0 .0 1) ;COX 2mRNA表达与患者性别、年龄及肿瘤大小、Lauren分型、分化程度均无显著性相关 (P >0 .0 5 ) ,而与肿瘤淋巴结转移、远处转移、临床分期和部位密切相关 (P <0 .0 1或P <0 .0 5 )。结论 :COX 2mRNA表达水平在人胃癌组织中增高 ,并与胃癌淋巴结转移、远处转移、分期、部位等临床病理特征密切相关 ,COX 2mRNA表达在人胃癌发生、发展。 Objective To test the expression of COX-2 gene in human gastric carcinoma, adjacent nontumorous tissues and normal gastric mucosa. Methods Samples of cancer tissues, adjacent nontumorous tissues and normal gastric mucosa from twenty-four patients with primary gastric cancer were obtained during surgery. COX-2 mRNA was measured by reverse transcription polymerase chain reaction (RT-PCR). Results The level of COX-2 mRNA in gastric carcinoma was significantly higher than that in adjacent nontumorous tissues and normal gastric mucosa (P< 0.01). There was no significant difference of the level of COX-2 mRNA between adjacent nontumorous tissues and normal gastric mucosa (P> 0.05). The level of COX-2 mRNA was not related to gender ( P> 0.05), age ( P> 0.05), tumor size ( P> 0.05), Lauren's classification ( P> 0.05) and differentiation degree ( P> 0.05), but was correlated with lymphnode metastasis ( P< 0.01), distant metastasis ( P< 0.01), clinical stage ( P< 0.05) and tumor location ( P< 0.05). Conclusion The expression of COX-2 gene is increased in gastric carcinoma, and correlates with tumor lymphnode metastasis, distant metastasis, clinical stage and tumor location. COX-2 plays an important role in the growth and development of gastric carcinoma.
