大肠癌组织中E-钙粘素α-环连蛋白和β-环连蛋白表达及其在生物学行为评价中的作用

摘要 为探讨E—Cad介导的粘附系统分子E-钙粘素(E—Cad),α-环连蛋白(α—Cat)和β-环连蛋白(β-Cat)在大肠癌中的表达及其与大肠癌生物学行为的关系,采用免疫组化S—P法对43例结直肠癌标本中E—Cad,α—Cat和β-Cat表达情况进行研究,分析其与临床病理因素间关系及其内在联系。E—Cad,α—Cat和β-Cat在大肠癌中表达分别为20.9%(9/43),60.5%(26/43)和37.2%(16/43)。E—Cad表达减低和肿瘤分化相关,与浸润深度、淋巴结转移和Dukes分期无关;α—Cat减低表达和肿瘤分化、浸润深度、淋巴结转移和Dukes分期相关。β-Cat表达和肿瘤分化、淋巴结转移相关,随浸润深度、Dukes分期升高而增加,但无统计学差异。E—Cad,α—Cat和β-cat共同阳性表达转移率12.5%(2/16)明显低于异常表达66.7%(18/27),各种异常表达模式间转移率无明显差异。结果表明,E—Cad和α—Cat,β-Cat复合物作为一个功能整体发挥细胞粘附作用,任何一种分子异常改变都易导致肿瘤发生转移,在这些异常改变中,α—Cat最常发生。E—Cad,α-Cat和β-Cat的表达对大肠癌生物学行为评估有重要意义,其预后价值有待研究。
