摘要 为了探讨维生素A(VA)对造血微环境调节的机理,采用体外培养和原位杂交技术动态检测全反式视黄酸(ATRA)诱导小鼠骨髓基质细胞(BMSC)30,60和120分钟后c-fos,c-jun及GM-CSF mRNA的表达。结果表明:①ATRA处理30分钟后c-fos和c-jun的表达即有增强,与对照组相比有显著差异(P<0.01),并持续表达至60分钟和120分钟时降至对照组水平;②GM-CSFmRNA的表达在30分钟和60分钟时与对照组水平相比无显著性差别,直至120分钟才表现出显著性差异(P<0.01)。实验结果提示:VA对造血微环境调节的机理可能是通过诱导BMSC中c-fos和c-jun mRNA的表达进而调控造血生长因子GM-CSF的分泌。 The study was designed to study the mechanism of the effect of all trans-retinoic acid (ATRA) on hematopoietic microenvironment. The bone marrow stromal cells were cultured in vitro, and in situ hybridization technique was used to examine the expression of proto-oncogene c-jun, c-fos and GM-CSF mRNA in mouse marrow stromal cells following exposure to the ATRA (10 mol/L). The expression of c-jun and c-fos mRNA was increased at 30 minutes and continued until a peak was reached at 60 minutes. Both c-jun and c-fos mRNA decreased and returned to control level at 120 minutes after exposure to ATRA. GM-CSF mRNA was no significantly difference as compared with control group until 120 minutes after ATRA stimulation. The results suggested that the mechanism of the regulation of retinoic acid on hematopoietic stromal cells might be induced the expression of proto-oncogene c-jun and c-fos in marrow stromal cells then the expression of GM-CSF.
