摘要
为了进一步阐明血小板生成素(thrombopoietin,Tpo)的生物学作用,我们研究了rhTpo(recombinant human Tpo)对人红白血病(human erythroleukemia,HEL)细胞增殖的影响。采用血浆凝块集落培养法和5-溴-2’-脱氧尿核苷(5-BrdU)掺入HEL细胞DNA-ELISA法。培养基中加入rhTpo,HEL细胞集落数量和吸光度值明显高于未加rhTPo对照组。当rhTPo浓度为10,50,100,200ng/ml时,实验组集落数分别是对照组集落数的1.5,2.07,2.34,2.46倍;实验组A值分别是对照组A值的1.28,1.84,2.25,2.31倍。研究结果表明,rhTpo促进HEL细胞增殖和DNA合成。
In order to further elucidate the biologic effects of Tpo (thrombopoietin), we have studied the action of rhTpo (recombinant human thrombopoietin) on the proliferation of HEL (human erythroleukemia) cells. The plasma clot assay and the ELISA of 5-BrdU incorporated into the DNA of HEL cells were used. In the medium treated with rhTpo 10, 50, 100, 200 ng/ml, the colonies of HEL cells and the absorbance (A405nm) increased as compared with control. The numbers of colonies in the experiment group respectively were 1.5, 2. 07, 2. 34, and 2.46 times as many as that in the control group, and the quantities of absorbance in the experiment group respectively were 1.28, 1.84, 2.25, and 2.31 times as many as that in the control group. These results show that rhTpo may enhance the proliferation and DNA synthesis of HEL cells.
