摘要
建立了一种电化学方法用于分析细胞凋亡和抗肿瘤药物的作用。应用特殊而简便的电极系统分析足叶乙甙(Vp-16)诱导的K562细胞凋亡。观察凋亡细胞的形态学改变和被动电化学行为。结果表明:DNA凝胶电泳和流式细胞仪分析均显示出典型的细胞凋亡特征,同时凋亡细胞的电化学伏安行为也明显不同,表现为凋亡细胞的峰值电流和电子转移速率的下降。结论提示电化学方法不仅能够反映细胞凋亡,尤其是早期细胞凋亡,而且提供了一种评价活细胞氧化还原系统改变的方法。
An electrochemical method for analyzing the apoptosis of K562 cells and the effectiveness of antitumor drugs has been developed. In this paper, the special but simple electrode system was used to study apoptosis of K562 cells induced by antitumor drug Vp-16. The morphological observation and the passive electrochemical behavior of K562 cells were studied for the time course of the cell culture with the drug. The analysis of DNA fragmentation by agarose gel electrophoresis and flow cytometry showed typical apoptotic result. At the same time, the electrochemical voltammetric behavior of the apoptotic K562 cells showed different from that of K562 cells with no drug added, such as the decreasing of peak current and electron transfer rate for the apoptotic cells. The comparison illustrates that the electrochemical method not only can reflect apoptosis, especially in early time, but also provide a method to evaluate the change in redox system of living cells as well.
