小麦赤霉病菌种保存及致病性研究 被引量:4

摘要 在小麦赤霉病抗性鉴定工作中,常因禾谷镰刀菌于室内连续多代移植培养导致菌株的致病性衰弱,而影响年度间鉴定结果的准确性和一致性。对此,笔者尝试"一次制种,多年应用",将小麦赤霉病菌制备成小米粒菌体,置4℃冰箱环境中长期保存。对保存年限不同的4个菌株进行室内培养和大田接种以测定其活力和致病性。结果表明,小麦赤霉菌小米粒菌体在4℃冰箱环境中可长期保存,7a之久仍有活力,且其田间致病性没有发生衰退。该菌种制作保存方法简单、经济、可靠,适用于小麦种质材料抗赤霉病鉴定选择工作。 Pathogen of wheat scab usually degenerates as Fusarium graminearum is successively transplanted and cultivated indoor during identification of resistance to wheat scab, this affects result of identification on accuracy and continuity in various years. To this appearance, authors attempted to make pathogen once and use several years. The pathogen was transplanted and cultivated on millet grain to form granulated fungi colonies. Then conserved under 4℃ in refrigerator for a long time. four strains being conserved in different time were used to determine vitality and pathogenicity by cultivating indoor and inoculating on farmland. Results indicated that pathogen of wheat scab on millet grain can conserve for long time under 4 ℃ in refrigerator and still keep vitality after 7 years the pathogenicity didn′t degenerate on farmland. The method of making and conserving pathogen is simple?economical and reliable, so it can be used for identification of resistance to wheat scab.
