摘要
目的 了解高三尖杉酯碱 (HHT)对鼻咽癌细胞CNE 2Z的caspase 3蛋白和mRNA表达的影响。方法 分别用HHT 0 (对照 )、0 12 5、0 2 5、0 5、1mg·L-1处理CNE 2Z细胞 8h ,采用WesternBlot分析caspase 3酶原蛋白表达的变化 ,用半定量RT PCR检测caspase 3mRNA表达的改变。结果 随HHT处理浓度的增加 ,caspase 3酶原的蛋白表达的减少具有统计学意义 (P <0 0 1) ,而caspase 3mRNA的表达的增高具有统计学意义 (P <0 0 1)。 0 2 5、0 5、1mg·L-1HHT处理组caspase 3酶原的蛋白表达均明显低于对照组 (P <0 0 1) ,1mg·L-1HHT处理组caspase 3mRNA的表达明显高于对照组(P <0 0 1)。结论 HHT可剂量依赖性地下调caspase 3酶原的蛋白表达 ,剂量依赖性地上调caspase 3mRNA的表达 ,HHT处理CNE 2Z细胞过程中可能有caspase
AIM To investigate the effects of homoharringtonine (HHT) on the expression of caspase-3 pro-tein and mRNA in nasopharyngeal carcinoma cells CNE-2Z. METHODS After CNE-2Z cells were treated with HHT [0(control),0.125,0.25,0.5,1 mg·L -1] for 8 h, the expression of pro-caspase-3 protein was analyzed by Western Blot and the expression of caspase-3 mRNA was detected by semi-quantitative RT-PCR. RESULTS As HHT-concentration increased, the expression of pro-caspase-3 protein decreased significantly (P<0.01) while the expression of caspase-3 mRNA increased statistically(P<0.01). The expression of pro-caspase-3 protein in HHT-treated at 0.25,0.5,1 mg·L -1 group was much higher than that in control group(P<0.01). The expression of caspase-3 mRNA in HHT-treated at 1 mg·L -1 group was obviously higher than that in control group (P<0.01). CONCLUSION HHT can down-regulate the expression of pro-caspase-3 protein and up-regulate the expression of caspase-3 mRNA in a dose-dependent manner. Caspase-3 may be activated during treatment with HHT in CNE-2Z cells.
