摘要
目的 :探讨Qi盐对尿酸 (UA)形成和排泄的影响。方法 :通过体外测定黄嘌呤氧化酶 (XO)抑制率来观察Qi盐对UA形成的影响 ;通过对高尿酸血症模型大鼠的血清和尿液UA含量的测定 ,以及大鼠酚红 (PSP)排泄试验来观察Qi盐对UA排泄的影响。结果 :①同等浓度下 ,Qi盐对XO的抑制百分率显然高于海盐 ;同时XO的抑制百分率随Qi盐的含量增加而增大 ;Qi盐的IC50 为 174 .7mg·L-1;阳性药物别嘌醇的IC50 为 1.0 7mg·L-1。②用氧嗪酸钾盐制备的高尿酸血症大鼠模型中 ,Qi盐大、中、小剂量 ( 1.6 ,0 .8,0 .4g·kg-1)组在 1~ 5h期间能显著降低血清UA水平 (P <0 .0 1或P <0 .0 5 ) ,且呈一定的量 效关系。Qi盐 3个剂量组和海盐组均能显著地增加高尿酸模型大鼠的排尿量和UA排泄总量 (P均 <0 .0 1)。③Qi盐 (在 0 .1~0 .4g·kg-1)对大鼠的PSP排泄试验 ,未观察到对肾小管重吸收和分泌功能的影响。结论 :Qi盐在体外具有一定的抑制XO的活性 ,能降低高尿酸血症模型大鼠的血清UA水平并可增加UA排泄量 ,且呈一定的量 效关系 ;
AIM:To study effects of Qi salt on the formation or excretion of uric acid. METHODS :①Studies on xanthine oxidase(XO) inhibitory activity of Qi salt in vitro ;②Effects of Qi salt on serum uric acid level and excretion in hyperuricemia rats model induced by potassium oxonate; ③A excretion test of phenolsulfonphthalein (PSP). By vitro and vivo experiments Qi salt has been evaluated for effects on the formation or excretion of uric acid. RESULTS: ①At the concentration of 100 mg·L -1 the inhibitory activities of Qi salt and sea salt on XO was 40.5% and 12.7% respectively. At the concentration of 50 mg·L -1 the inhibitory activities of Qi salt and sea salt on XO was 29.7% and 3.17% respectively. The IC 50 value of Qi salt was 174.7 mg·L -1 and the IC 50 value of allopurinol used as a positive control was 1.07 mg·L -1 . The dose effect relation was observed.② By treatment with Qi salt (1.6, 0.8, 0.4 g·kg -1 ) the serum uric acid level of hyperuricemia rats model pretreated with potassium oxonate was significantly decreased during 1 5 h ( P <0.01 or P <0.05). The dose effect relation was observed. From the data, the total amount of excretion of the urine and uric acid was significantly increased( P <0.01). ③In the test of PSP excretion no effect was observed for reabsortion and secretion of the renal proximal tubule by Qi salt(0 1-0 4 g·kg -1 ). It suggested that Qi salt possessed not uricosuric activity. CONCLUSION: The inhibitory activity of Qi salt on XO was observed in vitro . The serum uric acid level of hyperuricemia rats model was decreased significantly and the dose effect relation was observed by treatment with Qi salt. Uricosuric activity was not observed by treatment with Qi salt in excretion test of PSP.
