摘要
目的 :观察天花粉蛋白 (trichosanthin,TCS)在体外诱导 H2 2 肝癌细胞凋亡的效果。方法 :以不同剂量的 TCS加入到H2 2 肝癌细胞溶液中 ,在 2 4孔培养板上培养 72 h后荧光染色并涂片 ,激光共聚焦显微镜下观察 TCS诱导 H2 2 肝癌细胞凋亡的作用效果。结果 :10 μg/ml浓度的 TCS就可诱导 H2 2 肝癌细胞出现典型的凋亡形态学特征。经统计学分析 ,10 0 μg/ml TCS组与 5 0 μg/ml TCS组间差异无统计学意义 (P>0 .0 5 ) ,其余各组间差异均有统计学意义 (P<0 .0 1)。TCS的作用浓度与凋亡指数间有正相关关系 (r=0 .9713,P<0 .0 1)。结论 :TCS在体外可诱导 H2 2 肝癌细胞发生凋亡 ,诱导 H2 2 肝癌细胞凋亡是 TCS抗肝癌作用的机制之一 ,具有成为肝癌治疗新药的潜在价值。
Objective:To study the apoptosis-inducing effect of trichosanthin on hepatocarcinoma cell H 22 in vitro.Methods:A series of TCS with different concentrations were added to hepatocarcinoma cell H 22,then incubated for 72h in 24-well tissue culture microplates.All cells were detected and observed to estimate the effect of TCS by laser scanning confocal microscope.Results:The 10μg/ml TCS group exhibited morphologic features of apoptosis.There was a positive correlation between apoptosis and the concentration of TCS. (r=0.9713,P<0.01). Conclusion:TCS was able to induce apoptosis on hepatocarcinoma cell H 22.Apoptosis induced by TCS was one of the important mechanisms of the treatment of hepatocarcinoma.TCS might be promising in treatment of hepatocarcinoma and worthy of further study.
