摘要 目的 :探索应用非变性聚丙烯酰胺凝胶电泳分离大鼠海马组织蛋白的最佳条件。方法 :选用不同 p H的正极缓冲液和样品蛋白上样量 ,应用非变性聚丙烯酰胺凝胶电泳分离大鼠海马组织蛋白质。结果 :在其它条件不变的情况下 ,p H9.0的正极电泳缓冲液以及 40~ 6 0 μg/孔样品蛋白上样量时 ,可得到分辨率高、条带多而清晰的电泳图谱。结论 :应用 p H9.0的正极电泳缓冲液和样品上样量为 40~ 6 0 μg/孔时 ,进行非变性聚丙烯酰胺凝胶电泳 ,可成功地分离大鼠海马组织蛋白。 Objective:To find the optimum factors for separating proteins of hippocampi tissue in nondeture PAGE.Methods:Nondeture PAGE was used to separate proteins in different pH and loading quantity of protein sample.Result:On the condition of pH9.0 positive pole electrophoresis buffer and 40~60μg sample per well,electrophoresis map in high resolution was obtained,and the straps are distinct on the electrophoresis map.Conclusiont:Nondeture PAGE separated proteins of hippocampi tissue successfully on the best condition of pH9.0 positive pole electrophoresis buffer and 40~60μg sample per well.
