Determination of 5-Methoxytryptophol by Flow Injection Analysis with Chemiluminescence Detection

摘要 在甲醛的存在下 ,5 -甲氧基色醇 (5 - ML)与高锰酸钾在高氯酸介质中发生化学反应 ,能产生很强的化学发光。本文据此采用流动注射技术研究其发光行为 ,对影响化学发光强度的实验因素进行优化 ,建立了快速灵敏测定 5 - ML的化学发光新方法 ,测定方法的线性范围为 2 .0× 10 -8mol/ L— 4 .0× 10 -5mol/ L,检出限为 7.9× 10 -9mol/ L(S/ N=3) ,相对标准偏差为 2 .4 % (5 .7× 10 -7mol/ L5 - ML;n=10 )。该方法应用于脑脊液样品中 5 - ML含量测定 ,回收率达到 86 .0 %— 97.2 %。 A simple, rapid and sensitive method for the determination of 5-methoxytryptophol(5-ML) using flow injection analysis with chemiluminescence(CL) detection was developed based on the chemiluminescence reaction of acidic potassium permanganate and 5-ML with formaldehyde as an enhancer in perchloric acid medium.The experimental conditions for the method were optimized.The linear relationship between the chemiluminescence intensity and concentration of 5-ML is in the range of 2.0×10 -8mol/L-4.0×10 -5mol/L.The precision (measured as relative standard deviation) for 5-ML is 2.4% for 10 measurements at 5.7×10 -7mol/L standard solution,and the detection limits (signal-to-noise ratio 3) is 7.9×10 -9mol/L The recommended method was successfully applied to the determination of 5-ML in human cerebrospinal fluid(CSF), the analytical result is satisfactory with the recoveries in the range of 86.0%-97.2%.Singlet oxygen is suggested to be produced by the reaction and is responsible for the CL of 5-ML-KMnO 4-HCHO in HClO 4 solution.
