摘要
目的 探讨5α-还原酶在前列腺组织中的分布和表达情况。方法 应用免疫组化技术对6例正常前列腺、30例增生前列腺及20例前列腺癌组织中两型5α-还原酶的分布和表达情况进行研究。结果 前列腺组织的上皮和间质均有两型5α-还原酶的表达;前列腺增生组织中以Ⅱ型5α-还原酶的表达为主,且上皮中Ⅱ型5α-还原酶的表达强于间质;Ⅰ型5α-还原酶在前列腺癌上皮中的表达强于增生前列腺组织,两型5α-还原酶在前列腺癌上皮中的表达均强于间质。结论 双氢睾酮可能以自分泌和旁分泌两种方式作用于前列腺上皮;Ⅱ型5α-还原酶在前列腺增生中可能起主要作用,其在不同部位的不均匀分布反映了不同区域腺体增生速度和程度的差异。
Objective To investigate the distribution and expression of human 5α- reductase isoenzymes in prostate tissues. Methods From 2000 to 2002, the expression human 5α- reductase isoenzymes were examined by immunohistochemistry. Results Two isoenzymes in normal prostate(6 cases) , benign hyperplastic prostate (30cases) ,and prostate cancerous tissues(20cases) was detected in epithelial and stromal cells of prostatic tissues; In benign hyperplastic prostate tissues, type Ⅱ 5α - reductase was the predominant enzyme with the strongest staining,and the staining of type Ⅱ 5α- reductase in epithelial cells was stronger than that in stromal cells; in epithelial cells of cancerous tissues, type Ⅰ 5α- reductase had stronger staining than that in benign hyperplastic prostate tissues; in prostate cancerous tissues, the staining of both two type of 5α- reductases in epithelial cells was stronger than that in stromal cells. Conclusion Dihydrotestosterone may act on epithelial cells by means of autocrine and paracrine; type Ⅱ 5α -reductase may play major role in BPH; its uneven distributions at different areas reflect difference of velocity and extent of hyperplastic glands.
