中药复方对癫痫小鼠海马c-fos蛋白表达的影响 被引量:7

摘要 目的 探讨中药复方对戊四唑 (PTZ)致痫小鼠海马不同亚区c -fos蛋白表达的影响。方法 将 5 0只昆明种小鼠随机分为空白对照组、模型组、中药复方Ⅰ组、中药复方Ⅱ组及西药组。中药复方Ⅰ组和Ⅱ组灌服不同浓度的中药复方浓缩剂 ;空白对照组、模型组灌服蒸馏水 ;西药组灌服苯妥英钠。除空白对照组外 ,其余各组于第 6天给药 30min后腹腔注射PTZ( 5 5mL/kg)致痫 ,5 5min后处死动物。采用免疫组化法观察中药复方对癫痫小鼠海马不同亚区c-fos蛋白的影响。结果 PTZ致痫后c -fos表达增强 ,分布密集 ,染色加深 ;而中西药组表现为c -fos表达减少 ,分布稀疏 ,染色变浅。结论 中药复方能有效对抗PTZ所致的惊厥发作 ,可能与其能调节海马回内的c-fos蛋白的过度表达有关。 Objective It is to study the effect of Chinese herb compound on the expression of c-fos protein in hippocampus subareas of epileptic mouse induced with pentylenetetrazol (PTZ). Methods Fifty Kunming mice were randomly divided into normal control group, model group, Chinese herb compound Ⅰ group, Chinese herb compound Ⅱ group and western medicine group. Chinese herb compound Ⅰ group and Chinese herb compound Ⅱ group were administrated with different concentration of Chinese herb compound. Normal control group and model group were treated with distilled water. Western medicine group was treated with difetoin. On the sixth day, all of them, except the normal control group, were intraperitoneal injected with PTZ. 55 minutes later, all were sacrificed. The effects of Chinese herb compound on the expression of c-fos protein in hippocampus subareas of epileptic mouse were detected with immunohistochemistry method. Results The expression of c-fos protein, density and staining in model group increased, while those in western medicine group decreased. Conclusion Chinese herb compound can inhibit convulsion onset, whose mechanism is related to regulating of the c-fos protein's expression.
