Effects of IL-1 and PDGF on Secretion of MMP-9 in Vascular Smooth Muscle Cells

摘要 目的 :观察白介素 1和血小板源性生长因子对血管平滑肌细胞基质金属蛋白酶 9(MMP 9)表达的影响及其作用机制。方法 :用酶谱分析法检测MMP 9蛋白水平 ,采用电泳迁移率变动分析法测转录因子AP 1结合活性 ,用RT PCR法检测MMP 9的mRNA水平。结果 :白介素 1(2 0ng/ml)和血小板源性生长因子 (2 0ng/ml)联合使用显著增加MMP 9蛋白表达 ,提高AP 1结合活性 ,使MMP 9的mRNA水平明显增高。结论 :白介素 1和血小板源性生长因子联合使用可刺激MMP Objective: Our aim was to investigate effects of interleukin 1(IL 1) and plate derived growth factor(PDGF) on secretion of matrix metalloproteinase 9 (MMP 9) in vascular smooth muscle cells and its mechanism. Methods: Expression of MMP 9 was detected with zymography. The AP 1 binding activity was detected with electrophoretic mobility shift assay. The mRNA level of MMP 9 was detected with RT PCR. Results: Combination of IL 1α(20 ng/ml) and PDGF (20 ng/ml)significantly induced the secretion of MMP 9, increased the AP 1 binding activity and mRNA level of MMP 9. Conclusion: Combination of IL 1α and PDGF can induce expression of MMP 9.
