Merits and Demerits of Applying Western blot Technique in Diagnosing Trichinella Spiralis Infection

摘要 旋毛虫幼虫可溶性抗原(TSA)经SDS-PAGE和Western blot试验,用旋毛虫病人血清识别,显示18条蛋白染色带,分子量范围在80~30 kD之间,有7条清晰而明显的主带,分子量分别为80、74、66、63、55、42和40 kD,其中66、63和55 kD为旋毛虫特异性的蛋白带。TSA与日本血吸虫病人血清抗体应答有11条染色带,分子量范围在42~25 kD之间。根据蛋白染色带分子量的差异,可以进行两种病的鉴别诊断,与正常人和其它8种寄生虫病人血清反应均未见清晰的蛋白染色带。 Soluble larve antigen of Trichinella spiralis was analysed by SDS-PAGE and Western blot. Preliminary examinations with pooled human sera taken from 39 cases of trichinosis rewealed 18 protein bands, and their molecular weights rangen from 80 to 30 kD. The molec- ular weight of the most prominent protein bands was 80,74,66,63,55,42 and 40 kD. A- mong them, only those with molecular weight of 66,63 and 55 kD were likely to be specific to Trictinella spiralis and of diagnostic value. The reactions of this soluble antigen with pooled sera of 35 cases of Schistosomiasis japonica showed 11 protein bands and their molecu- lar weights ranged from 42 to 25 kD. Sixty sera of normal individuals and 75 sera of patients with parasitic diseases other than Trichinosis all gave negative reactions. Thus Trichinellosis may be differentiated from Schistosomiasis by their diffences in protein bands.
