摘要
将日本血吸虫虫卵可溶性抗原点加于微孔滤膜 ,经预作用和封闭后 ,依次加入待检血清和金标记二抗 ,建立快速、敏感、特异性强 ,操作简便和经济实用的诊断日本血吸虫病的免疫学检测方法———快速斑点免疫金染色法 (R Dot IGS) ,该法无需银显影 ,检测过程仅为 15min。用 (R Dot IGS)、快速微量斑点免疫金银染色法 (RM Dot IGSS)和斑点免疫金银染色法 (Dot IGSS)同步检测 5 5份日本血吸虫病患者血清 ,三种方法的阳性率分别为 98. 18% ( 5 4/ 5 5 ) ,98. 18% ( 5 4/ 5 5 )和 10 0 % ;检测 5 0例健康人血清均为阴性 ;检测 10例肝吸虫病人和 10例肺吸虫病人血清 ,仅有 1份肝吸虫病人血清在RM Dot IGSS反应为阳性。检测结果证明R Dot IGS具有敏感特异、快速简便实用的特点 ,适用于日本血吸虫病的现场流行病学调查和临床快速诊断。
A rapid, specific, sensitive, convenient and economical immunological assay,rapid-dot-immunogold staining( R-Dot-IGS),was set up for detecting sera antibodies of the patients infected with Schistosoma japonicum. The soluble egg antigens of Schistosoma japonicum were added onto micropore filter membrane. After pre-reacting and blocking, the sample serum to be detected and sheep anti-human IgG labelled with chloroauric acid were added sequentially to read the results. The assay only took about 15 minutes. As controls, the Dot-immunogold silver staining (Dot-IGSS) and rapid micro volume Dot-IGSS (RM-Dot-IGSS) were performed simultaneously. The positive rate of the sera, which were collected from 55 confirmed cases with Schistosomiasis japonica, were 98.18%, 98.18%, 100%, when detected by the R-Dot-IGS, RM Dot-IGSS and Dot-IGSS respectively. The sera of 50 healthy controls, 10 cases of clonorchiasis and 10 cases of paragonimiasis, all showed negative reactions, except one case with clonorchiasis showed positive reaction with RM-Dot-IGSS. Compared with Dot-IGSS and RM Dot-IGSS,R-Dot-IGS has similar sensitivity and specificity, but the latter is more rapid, simpler and more economical, and hence is worth for application in field situations and clinical rapid diagnosis of Schistosomiasis japonica.
