用South-Western blot mapping方法观察锥虫VSG基因转录酶与基因组核酸片段的结合

摘要 应用South—Western blot mapping方法观察了初步提纯的锥虫转录变异抗原基因的聚合酶与其基因组DNA片段的结合情况.在选定的试验条件下,初步纯化的RNA聚合酶既能与变异抗原基因结合,也能与一些待鉴定的非特异性核酸片段结合.本文对存在的问题进行了讨论. The interaction of transcriptase for variant surface glycoprotein (VSG) genes with genomic DNA of trypanosomes was examined by South-western blot mapping. The bound DNA was eluted from protein-DNA complex and analyzed by agarose gel electrophoresis. The results indicated that the primarily purified VSG transcriptase, could combine with VSG genes and some other DNA fragments. Possibility for improvement of sequence-specific binding was discussed.
