IFN-α影响淋巴细胞迁移至小鼠皮肤区的实验研究

Effects of Interferon-α on spleen lymphocyte migration in mouse skin

目的 体内观察IFN α对淋巴细胞迁移至小鼠皮肤区的影响。方法 以近交系BALB/C小鼠为实验动物 ,皮内注射IFN α ,对照组皮内注射生理盐水 (NS) ,同时尾静脉输注3H -UR标记的淋巴细胞 ,然后于不同时间切取注射区皮块 ,制样后行乳化计数测量。测量值经计算后转化成细胞浓度因子(CCF) ,用CCF值反应淋巴细胞迁移的水平。结果 不同剂量的IFN α注射区CCF值均高于对照组 ;注射IFN α后不同时间检测显示 ,6h时CCF值最大 ,至 2 4h时逐渐减小。结论 IFN α可以体内诱导淋巴细胞迁移至皮肤区 ,并且呈现剂量依赖性关系 ,在 6h时作用最强 ,以后至 2

Objective To examine the effect of IFN α on lym ph ocyte migration in mouse skin.Methods Female BALB/C mice w ere divided randomly into IFN α and NS groups.The skins on the back of all mic e were shaved,0 05ml of IFN α was injected intradermally.At the same time,spl een lymphocytes labeled with 3 H UR were Ⅰ.Ⅴ injected.Different time later, the injected areas were punched out,throung emulsion counting,the counts per min ute(CPM)of 3 H was examined,and the cell concentration factor(CCF) was determin ed by calculation. Results The CCF of IFN α groups were high er than those of NS groups,kinetic analysis shows that the maximun response to I FN α was observed at 6h,gradually decreased till to 24h.Conclusion IFN α can stimulate lymphocytes migration to skin in vivo in dose depen dent fashion.
