摘要
研究了AFLP标记在乌羊遗传多态性方面应用的可行性和该山羊个体基因组DNA的AFLP扩散结果。实验应用10条人工设计的与接头序列相识别的AFLP选择性引物,用PstⅠ酶切,对15只乌羊基因组DNA进行AFLP反应,共获得116个AFLP标记,单引物获得的标记数在2～21之间,乌羊群体相似系数AFLP研究结果为0.897(0.798～0.976),遗传距离为0.024～0.202之间。该研究为评价乌羊的遗传稳定性提供了相关的参数,准确评价尚待和其它品种对比研究后确定。
We studied the application of AFLP markers to detect genetic polymorphic loci in wu goats and their amplified AFLP results.Their genetic construction of the population was also analysed.10AFLP primers were used.genetic DNA from15goats were restrictive by PstⅠ,116AFLP markers were obtained,obtaining the markers of individual primer were between2and21.The results indicated that the similary index of population in miniature wu goats was0.897(0.798to0.976),genetic distance was from0.024to0.202AFLP technique was an effective method in genetic polymorphism monitoring of wu goats,supplying useful parameter with appraise genetic stability of wu goats.
