杏果实苹果酸脱氢酶基因克隆及定量表达分析 被引量:1

摘要 以新世纪杏(Armeniaca L.cv.Xinshiji)为试材,采用RT-PCR方法,克隆了果实中苹果酸脱氢酶的编码基因,并对其在不同时期的表达进行了定量检测。序列分析表明:苹果酸脱氢酶基因的近全长cDNA长度为1081bp,它与Gen Bank中登记号为AF367442.1的桃子的苹果酸脱氢酶基因高度同源,核苷酸序列同源性为99%;该基因已在Gen Bank中注册,登记号为HQ189434。苹果酸脱氢酶基因的表达量在果实发育前期最高,随着果实的发育表达量逐渐降低,在果实发育后期表达量略有上升,其变化趋势与苹果酸脱氢酶的活性变化规律相一致,但基因的表达量较酶活性变化幅度小。 MDH Genes was cloned from the Xinshiji apricot fruit( Armeniaca L. cv. Xinshiji),Investigation of Expression of MDH genes using Real- Time Fluorescence Quantitative RT- PCR. The result of testing sequence showed that the nucleotide sequence of the MDH gene was 1081 bp,and it shared 99%homology in nucleotide sequence with peach MDH gene,whose access number is AF367442. 1. The gene chit has been registered in Gen Bank with accession number HQ189434. During the fruit development,the expression of the MDH gene showed a decreasing tendency and it was increased the lowest on the 51 days after flowering and then went up slightly. The expression of the MDH gene and activity of the MDH showed identical.
