摘要
目的 :探讨肾缺血再灌注损伤时 ,小管上皮细胞ATP酶活性与自由基代谢的变化。方法 :利用离体培养小管细胞的缺血再灌注模型 ,观察ATP酶与超氧化物歧化酶 (superoxidedismutase ,SOD)、脂质过氧化产物丙二醛(malondialdehyde,MDA)的变化。结果 :缺血再灌注损伤导致肾小管细胞ATPase、SOD活性明显降低 ,而MDA含量明显增高 ,中药参麦注射液 (ShenMaiinjection ,SMI)能明显提高ATPase、SOD活性 ,同时脂质过氧化MDA含量明显降低。结论 :实验结果提示在缺血再灌注中 ,氧自由基损伤了细胞的ATP酶和脂质成分 。
Objective:To investigate the changes of the metabolism of free radical and ATPase activity during ischemia-reperfusion. Methods: The content of the lipid peroxidation product--malondialdehyde(MDA), and the activity of superoxide dismutase(SOD) and ATPase were detected in cultured tubular cellular model of renal ischemia-reperfusion injury. Results: Ischemia and reperfusion reduced SOD and ATPase activity while MDA increased significantly; these changes became less conspicuous in those cells treated with SMI at the same time. Conclusion: The findings indicated that free radical play a crucial role in the injury to ATPase during the reperfusion of ischemic renal, which could be inhibited by SMI.
