应用免疫酶组化技术检测狂犬病毒的方法学探讨 被引量:2

摘要 目的:比较常规石蜡切片HE染色和免疫酶组化ABC法检测狂犬病毒的灵敏度。方法:在已感染狂犬病毒的10只小鼠脑组织石蜡切片上,分别用HE染色和免疫酶组化ABC法检测狂犬病毒。结果:HE染色仅在4只小鼠脑组织神经细胞胞浆内找到代表狂犬病毒抗原的内基氏小体,阳性率为40%;免疫酶组化ABC法检测10只小鼠脑组织均见内基氏小体,其阳性率为100%,明显高于HE染色法(P<0.05)。结论:用免疫酶组化ABC法检测狂犬病毒抗原比HE染色法未列举特异的事实敏感,提示用免疫酶组化ABC法检测可提高狂犬病毒诊断率。 Objective:To compare which staining is more sensitivity between hematoxyin-eosin(HE)staining and immunoenzyme histochemistry ABC(ABC-IHC)on detecting Rabies virus.Method:10mice were infected by Rabies virus.The brain tissues on paraffin slides from the10in-fected mice were detected by both staining methods of HE and ABC-IHC.Results:The Negri body,which is taken as the sign of Rabies virus in neuron,was detected in4of10cases(40%)by HE staining,while in10of10cases(100%)by ABC-IHC staining.The positive rate got by ABC-IHC was much higher than that by HE(P<0.05).Conclusion:ABC-IHC staining method is more sensi-tive on detecting Rabies virus than HE staining method.
