Prokaryotic Expression and Identification of Human Spermidine/Spermine N^1-acetyltransferase

用RT—PCR从人肺癌细胞株中获得精脒/精胺N^1-乙酰基转移酶(SSAT)编码区cD—NA,克隆到大肠杆菌原核表达载体pET15b中,并让该载体在大肠杆菌中高效表达出SSAT重组蛋白,利用Ni—NTA树脂亲和层析纯化出重组蛋白,从而建立了SSAT原核表达和纯化系统。

A expression-purification system of human spermidine/spermine N^1-acetyltransferase(SSAT) was built by amplifying the cDNA of SSAT with RT-PCR from human lung cancer cells line and then cloned SSAT gene into a prokaryotic expression vector.The SSAT gene in E.coli was qualifiedly ex- pressed and the recombination protein was purified by Ni-NTA resin affinity chromatography.
