摘要
目的 从人胚胎心室肌细胞中提出了一种在体外可刺激Wistar小鼠原代心肌细胞DNA合成并具有生物活性的纯化物质 ,称之为心肌生长刺激因子 (MyocardiumGrowthStimulatingFactor,MGSF)。 方法 采用硫酸铵分级离法、阴离子纤维素交换层析、高压液相色谱洗脱、SDS -PAGE、毛细管电泳、高压液相色谱分离及生化鉴定。结果 该物质分子量为 3.7KDa ,含有 17种氨基酸 ,同最新的蛋白质氨基酸序列文库扫描进行同源性分析 ,并确定此序列可能为一种新的氨基酸序列 ;并对其进行了生化鉴定 ,其临床意义正在进一步研究这中。结论 该研究结果初步表明 ,MGSF可能为急性缺血性心脏病提供有效的药物治疗开辟了一条新的思路。
Objective:To separate and purify a protein-Myocardium Growth Factor(MGSF) that has bioactivity in vitro from ventricle muscle of human embryo.Methods:DEAE-52 cellulose exchange chromatography, SDS-PAGE and so on were used to separate and purify MGSF.And the purity, amino acid proximate analysis, N-terminal amino acid sequence of this protein and its bioactivity were studied by SDS-PAGE, capillary zone electrophoresis.Results:MGSF's molecular weight was about 3.7 Kda, it contained 17 kinds of amino acid, the homogenous material had been not found by scanning of protein library.Conclusions:MGSF may be a new drug to treat acute ischemic heart disease. And its clinical use should be further studied.
