摘要
目的 观察严重烫伤后巨噬细胞膜蛋白GTP酶活性变化及补体对它的影响 ,探讨它在烧伤后巨噬细胞活化中的作用。方法 采用小鼠 18%TBSAIII度烫伤模型 ,分为正常组、去补体对照组、各时相烫伤组以及用眼镜蛇毒素因子预先体内耗竭补体再烫伤组 ,取腹腔巨噬细胞 ,分离膜蛋白 ,以r 32p GTP为反应底物 ,通过液相闪烁计数检测其GTP酶活性。结果 腹腔巨噬细胞膜蛋白GTP酶活性在严重烫伤后 6h开始升高 ,72h仍处于高峰 ;去补体后再烫伤 ,则GTP酶活性显著低于烫伤对照组。
Objective To study the dynamic changes of GTPase activity of mouse peritoneal macrophage(PMф)after severe burn and the effects on it of activated complement, so as to explore the role of activated complement on G-protein activation. Methods The mice inflicted with 18% TBSA III degree scalding were employed as the model and randomly divided into normal control group, decomplementizing control group, scalding group and decomplementizing scalding group.The activity of membrane protein of PMф was examined by means of liquid scintilltion counting with r 32p GTP. Results The activity of GTPase increased 6 h after injury and persisted in the high level after 72 h.When complement was depleted with cobra venom factor(CVF),the GTPase activity decreased obviously compared with the burn control group. Conclusion The results indicate that G protein of PMф membrane is activated by the activated complement.
