柚杂交胚离体培养再生植株的研究 被引量:1

摘要 以王官溪蜜柚和度尾文旦柚互交产生的杂交种幼胚为材料,研究离体胚再生植株的技术.结果表明:幼胚在MT培养基上直接成苗质量最好;子叶在MT附加0.25mg·L-16-BA+0.5mg·L-1ZT+0.01mg·L-1NAA固体培养基中产生不定芽;带子叶胚轴在MT附加1mg·L-16-BA+0.2mg·L-1IAA+1mg·L-1GA3的固体培养基中可提高其萌芽率,繁殖系数扩大了20倍以上;分化的丛生芽是试管微芽嫁接的良好接穗材料,将芽苗转移到1/2MS+0.5mg·L-1IBA+0.5mg·L-1NAA的固体培养基中诱导生根壮苗最好. The technique of plant in vitro regeneration from young hybrid embryos between Guanxi Pummelo and Duwei Pomelo was developed. The results showed that seedlings directly generated from young embryos were the strongest on the solid MT medium. The adventitious buds were germinated from the cotyledon on the solid MT medium supplemented with 0.25 mg·L^(-1) 6-BA, 0.5 mg·L^(-1) ZT and 0.01 mg·L^(-1) NAA. The cotyledon axis was induced to outgrow differentiation shoots on solid MT medium with 1 mg·L^(-1) 6-BA, 0.2 mg·L^(-1) IAA and 1 mg·L^(-1) GA_3 and the germination rate was raised, the propagation coefficiets were increased more than 20 times. The differentiation buds were good materials of grafting for minute buds in vitro. It was recommend to transfer the shoots onto the solid 1/2MS medium with 0.5 mg·L^(-1) NAA and 0.5 mg·L^(-1) IBA for rooting and plantlet growth.
