摘要
用果胶酶和纤维素酶酶解的水仙根尖制备染色体标本,从大量中期分裂相中选择分散良好的染色体进行拍照和组型分析,建立核型图.根据核型识别目标染色体,应用显微操作系统成功进行单染色体分离.
The chromosome specimens were made from root tip dissociated with the mixture of pectase and celluase. The standard karyotype of narcissus was established based on many metaphase photos. The target chromosome was identified and microdissected using microglassneedles under the micromanipulator, then transferred into Eppendorf tube for further study. This is a successful way to microdissect the target chromosome in narcissus.
