摘要
用毛细管气相色谱研究了胆固醇及其8种氧化产物的分析方法。以5α-胆甾烷为内标物,经N,O-双(三甲基硅烷基)三氟乙酰胺(含1%三甲基氯硅烷)衍生化,在恒温条件下SE-30色谱柱上进行分离,用氢火焰离子化检测器(FID)检测。胆固醇及8种胆固醇氧化物的质量浓度在0.03-0.60g/L范围内与峰面积线性关系良好。对于不同组分的检测限在5.4-27ng之间。该法适用于常见胆固醇氧化物的分析,对花生油存在下胆固醇加热氧化情况进行了初步测定。
A method to analyze cholesterol and 8 cholesterol oxides was studied by capillary gas chromatogrphy.The internal standard was 5(-cholestane. N,O-bis(trimethylsilyl)trifluoroacetamide(1% trimethylchlorosilane) was used for derivative reagent in pyridine,The separation and analysis was performed on SE-30 column with a flame ionization detector under isothermal condition.The result demonstrated a good linear correlation between the FID response and the concentrations of components in the range 0.03 -0.60g/l,the limits of detection of chlolesterol oxides varied from 5.4 ng to 27 ng,depending on the compound.
