^(99m)Tc标记的人成骨肉瘤单克隆抗体在荷瘤裸鼠体内的分布及显像研究

摘要 目的 研究骨肉瘤的放射免疫显像。方法 应用杂交瘤技术获得OS 732骨肉瘤单克隆抗体 ,用99mTc直接标记单克隆抗体 (McAb) ,将99mTc McAb经尾静脉注入荷人成骨肉瘤裸鼠体内 ,然后进行SPECT显像。结果 注射后6h ,肿瘤影像模糊 ,2 4h后 ,肿瘤部位可见明显的放射性浓聚 ,而对照组99mTc IgG除在肝脏稍有积聚外 ,放射性分布弥散 ,未见肿瘤显像。注射99mTc McAb后 2 4h ,肿瘤中的放射性明显高于其他组织 ,此时肿瘤与血液的放射性之比为 2 .2 5 ,而对照组仅为 0 .97,故有明显差别。结论 99mTc McAb在骨肉瘤组织中有明显放射性浓聚 ,因此 ,99mTc McAb放射性显像对骨肉瘤及其转移灶有很好的诊断价值。 Objective To study the radioimmunoimaging of osteogenic sarcoma. Methods Acquire the monoclonal antibodies of anti-soteosarcoma cell line OS-732 with the technique of hybridoma and label the McAb with 99m Tc directly. Inject the McAb into the tail vein of the nude mice bearing human osteosarcoma then acquire the image with SPECT.Results 6 hours after injection, the tumors were visualized vaguely and after 24 hours the tumors were visualized obviously .But in the control group only the liver show a little higher radioactivity and the image of the tumors were not visualized. 24 hours after injecting, the radioactivity of the osteosarcoma is much higher than that of other tissues. The radioactivity ratio of osteosarcoma to the blood is 2.25, but the ratio in the control group is only 0.97. They are obviously different.Conclusion The radioactivity of the osteosarcoma is much higher than other tissues, so the radioimmunoimaging with the 99m Tc-McAb is very valuable in the diagnosis of the osteosarcoma and its metastases.
