摘要
建立快速预测铁皮石斛不同部位中的多糖、石斛碱、总黄酮含量的方法,以期为铁皮石斛茎、叶、花质量快速预测评价提供依据。采用红外光谱一维谱图及二阶导数宏观指纹鉴定的方法对铁皮石斛的花、茎、叶的红外光谱图进行了整体的分析,依据吸收峰强度预测铁皮石斛不同部位中活性成分含量。结果显示:铁皮石斛不同部位样品红外光谱近似度较高,吸收峰明显且较多,1 750~400 cm-1波段内吸收峰比较密集。铁皮石斛茎中的多糖成分含量最高,叶中含量次之,花中含量最少,多糖成分含量差别显著;铁皮石斛茎中的石斛碱含量最高,叶中含量次之,花中含量最低,石斛碱的含量差异不显著;铁皮石斛叶中的黄酮类物质含量最高,花中次之,茎中最低,总黄酮类含量差异显著;傅里叶变换红外光谱能实现铁皮石斛不同部位中多糖、石斛碱、总黄酮含量准确预测,为铁皮石斛不同生药部位质量评价提供快速、有效的方法。
A rapid method is developed for determination of polysaccharide,dendrobium alkaloid and total flavonoids in different parts of Dendrobium catenatum in order to provide the basis for the rapid evaluation of stem,leaf and flower of Dendrobium catenatum.Flower,stem and leaf of Dendrobium catenatum are analyzed by one-dimensional infrared spectra and second derivative fingerprint.Active component content in different parts of Dendrobium catenatum is predicted by absorption peak strength.The results show IR spectra of different parts of Dendrobium catenatum are similar to each other.The absorption peaks of IR spectra are numerous,and relatively dense in band 1750-400 cm-1.The content of polysaccharide in stem of Dendrobium catenatum is the highest,leaf is the next and flower is the least.In general,the content of polysaccharides(glycosides)in different parts is significantly different.Dendrobium catenatum has the highest content of dendrobium alkaloid in stem,the second highest in leaf and the lowest in flower.There is no significant difference in dendrobium alkaloid content.The content of flavonoids in the leaf of Dendrobium catenatum is the highest,followed by the flowers and the lowest in the stems.The content of total flavonoids in different parts is significantly different.The content of polysaccharides,dendrobium alkaloids and total flavonoids in different parts of dendrobium candidum can be predicted accurately by FTIR,which provides a rapid and effective method for the quality evaluation of different parts of dendrobium candidum.
