摘要
把保藏的144株枯草杆菌分成6个试验菌株组,用分羣混合双层法,从6个菌株组內分离到4株带噬菌体的菌株。选择其中一株,其所带的噬菌体对Ki-2菌株能转导的Bs47菌株,对其溶源性作了初步的观察。根据单菌落传代孢子加热以消除游离噬菌体后,仍继续释放噬菌体,以及Bs47菌株对其本身所释放的噬菌体表现免疫性等这些特点,从而确定Bs47是溶源性菌株。应用丝裂霉素c或紫外线进行产生噬菌体诱导试验,结果都有诱导效应。增加噬菌体产生量,前者达1,000倍,后者在10倍以上。
Four phage-carrying strains of Bacillus subtiliswere isolated by group-mixture test method from144 strains kept in this laboratory.Strain Bs47,owing to the transducing capabilitis of its phagc,was selected for further investigation.After serialisolation of single colony or heat treatment of thespores to get rid of the free phage,the bacteriawere still capable of releasing phage.The bacteriaalso showed complete resistance against infectionwith the phage released by themselves.Based onthese characteristics,B.subtilis strain 47 (Bs47) wasconsidered to be a typical lysogenic strain.Treat-ment with Mitomycin C or irradiated with ultra-violet light resulted in different degrees of increaseof phage production,and the effect of mitomycin cinduction was about 100 times higher than that ofultraviolet induction,or 1000 times higher than theuninduced.
