摘要
【目的】探讨甘草酸二铵在抑制炎症方面对脑缺血再灌注损伤的保护作用。【方法】42只雄性SD大鼠随机分为正常组(A组,n=6)、大脑中动脉闭塞再灌注(middle cerebral artery occlusion reperfusion,MCAOR)+生理盐水组(B组,n=15)、MCAOR+甘草酸二铵组(C组,n=15)和MCAOR后对照观察组(D组,n=6),Zea-Longa评分法评估神经功能,3 d后处死大鼠,Western blotting法检测p-38丝裂原活化蛋白激酶(p-38 mitogen-activated protein kinases,p-38MAPK)表达,免疫组化染色法检测核因子κB(nuclear factor kappa-B,NF-κB)和基质金属蛋白酶-9(matrix metalloprotein-9,MMP-9)表达情况。【结果】造模后神经功能明显恶化;3 d后,Zea-Longa评分、p-38MAPK表达、NF-κB和MMP-9免疫组化染色结果均显示B组与C组和D组比较有显著统计学差异(P<0.05)。【结论】甘草酸二铵能明显改善大鼠缺血再灌注损伤后神经功能状态,并能减少p-38MAPK和NF-κB炎症通路表达,并降低MMP-9炎症因子从而缓解大鼠缺血再灌注损伤。
【Objective】To explore the effect of diammonium glycyrrhizinate on inhibiting inflammation after cerebral ischemia reperfusion injury.【Methods】42 male SD rats were randomly divided into the normal group(group A,n=6),the middle cerebral artery occlusion reperfusion(MCAOR)+saline group(group B,n=15),the MCAOR+diammoniumglycyrrhizinate(DG)group(group C,n=15)and the MCAOR control group(group D,n=6).The neurological function was assessed by Zea-Longa scoring method.All the rats were sacrificed after 3 days.The expression of p-38 mitogen-activated protein kinases(p-38 MAPK)was detected by Western blotting.Immunohistochemical staining was used to detect the expression of nuclear factor kappa-B(NF-κB)and matrix metalloprotein-9(MMP-9).【Results】Neurological function deteriorated significantly after MACOR.The results showed that there was a significant statistical difference in the Zea-Longa score,and expression of p-38 MAPK,NF-κB and MMP-9 of group C and group D compared with group B(P<0.05).【Conclusion】DG can significantly improve the nerve function of rats after ischemia reperfusion injury,and reduce the expression of p-38 MAPK and NF-κB in inflammatory pathways,and decrease the expression of inflammatory cytokines MMP-9.Therefore,it can alleviate ischemic reperfusion injury.
