摘要
【目的】研究锌指转录因子ZFP580对大鼠星形胶质细胞中活性依赖的神经保护蛋白(activity-dependent neuroprotective protein,ADNP)表达的影响。【方法】培养大鼠星形胶质细胞,分别以不同浓度TGF-β1和SB431542刺激细胞一定时间后,RTPCR、Western blotting检测ZFP580和ADNP的表达。再以同一浓度TGF-β1和SB431542刺激细胞不同时间后,RT-PCR、Western blotting检测ZFP580和ADNP的表达。【结果】不同浓度TGF-β1刺激4 h后,ZFP580和ADNP mRNA和蛋白的表达均随TGF-β1浓度的升高而上升;同一浓度TGF-β1刺激细胞不同时间后,ZFP580和ADNP mRNA和蛋白的表达呈先上升后下降的趋势;不同浓度SB431542刺激细胞6 h后,ZFP580和ADNP mRNA和蛋白的表达均随SB431542浓度的升高而降低;同一浓度SB431542刺激细胞不同时间后,ZFP580和ADNP mRNA和蛋白的表达总体呈逐渐下降趋势。【结论】转录因子ZFP580在调节神经胶质细胞ADNP的表达中具有重要作用,其机制可能通过活化了TGF-β1/ALK5/Smad2/3信号通路。
【Objective】To reveal the effects of transcription factor ZFP580 on the expression of activity-dependent neuroprotective protein(ADNP)in rat astrocytes.【Methods】Rat astrocytes were cultured regularly.The cells were treated with different concentrations of TGF-β1 and SB431542 for some time and the same concentration of TGF-β1 and SB431542 for different time.The expressions of ZFP580 and ADNP were detected by RT-PCR and Western blotting.【Results】After 4-hour stimulation of TGF-β1 in different concentrations,the mRNA and protein expressions of ZFP580 and ADNP increased with the concentration.After the stimulation of TGF-β1 in the same concentration for different time,the expressions of ZFP580 and ADNP first increased and then decreased.After 6-hour stimulation of SB431542 in different concentrations,the mRNA and protein expressions of ZFP580 and ADNP decreased with the increase of concentrations.After the stimulation of SB431542 in the same concentration for different time,the mRNA and protein expressions of ZFP580 and ADNP decreased.【Conclusion】Transcription factor ZFP580 plays an important role in regulating the expression of ADNP in astrocytes.The mechanism may be involved in the activation of TGF-β1/ALK5/Smad2/3 signal pathway.
