摘要
目的 研究氟对血管平滑肌细胞增殖活力的影响。方法 对大鼠胸主动脉平滑肌细胞采用组织块培养法。应用四甲基偶氮唑蓝 (MTT)法测定细胞增殖活性和生化方法检测细胞内蛋白和丙二醛 (MDA)含量。结果 加氟培养胸主动脉平滑肌细胞 48、72h时间段在 1.0~ 15 .0mgF-/L组随着投氟量的增加 ,细胞增殖活性有所降低 ,且在 2 5 .0mgF-/L组各时间段细胞增殖活性降低最为明显。在同一时间段MDA含量随着投氟浓度的增加而增加 ,72h时间段的MDA含量较 2 4h时间段的相同氟作用浓度下MDA明显下降。结论 随着氟作用时间的延长和浓度的增加对血管平滑肌细胞增殖活性降低显著 。
Objective To study the toxicity of fluoride on vascular smooth muscle cell proliferation. Methods Utilizing MTT and biochemical methods to test the proliferationand the level of vascular smooth muscle cell protein and MDA. Results The cell proliferation is lower when the longer time and the higher content fluoride treated the vascular smooth muscle cell . The MDA showed lower in the vascular smooth muscle cells treated by fluoride 72 hour than that of 24 hour. Conclusions Fluoride did obvious harm to vascular smooth muscle cells proliferation while its content and time increased; oxidative stress possibly gets involved in the toxic mechanism of high fluoride.
